Applications of Fluorescein

Fluorescein is a common fluochrome dye; a yellowish to red crystalline powder; insoluble in water; soluble in dilute acids, and dilute alkali. It is obtained from the condensation of phthalic anhydride with resorcin on heating. It is used in dying silk and wool. It is the parent compound of eosin. All eosins are bromine derivatives of fluorescin, used in dyeing textiles, ink manufacturing, in coloring cosmetics, in coloring gasoline and as a toner. Eosin is a composition of Wright stain, in modified Nocht's stain and eosin-methylene blue medium. The sodium salt of fluorescein is called uranine which are used in dying silk and wool, in oceanography as a marker in sea water, and in coloring agrochemicals, fertilizers, detergents and antifreezers. Fluorescein sodium (uranine) is an orange-red crystalline powder; odorless, water-soluble.

In biology, it is used as a detection agent for visible images of structures or details which are not seen by eyes. It is used as a diagnostic indicator in assessing corneal trauma and fitting contact lenses. It is used retinal angiography detect latent blood stains for the study of antigen-antibody reactions in serum. The isothiocyanate salt of fluorescein is capable of being conjugated to amine groups of proteins inside cells, particularly antibodies, and hence used to label and track the molecules in various assays.
